The firebrat, Thermobia domestica (Packard) (Thysanura: Lepismatidae), aggregates in response to the faeces of conspecifics. This aggregation response is mediated by two microbial symbionts, the bacterium Enterobacter cloacae (Jordan) Hormaeche & Edwards (Enterobacteriaceae) and the fungus Mycotypha microspora Fenner (Mucorales). Our objective was to determine how these microbes are transmitted between firebrats. We produced fluorescently labelled E. cloacae and M. microspora and presented them to firebrats. Firebrats consumed large quantities of these labelled microbes and deposited them with their faeces where they proliferated rapidly. Firebrats did not harbour E. cloacae or M. microspora within their ovarioles or eggs, and thus cannot transmit them transovarially. Instead, firebrats acquired them horizontally whenever they fed on microbe‐contaminated material, such as faeces, faeces‐contaminated paper, or egg surfaces. Firebrats moult throughout their life, and with each moult they shed the cuticular lining of their digestive tract and likely any microbes residing therein. Because firebrats remain in close contact and live in groups of mixed age and gender, newly moulted individuals can readily re‐acquire E. cloacae or M. microspora from group members. Synanthedon myopaeformis Borkhausen (Lepidoptera: Sesiidae) is a diurnal clearwing moth native to Eurasia that was recently introduced into British Columbia (BC) and Ontario, Canada, where it has become a serious pest in apple orchards. In BC, these moths commonly feed on nectar of inflorescences, particularly that of showy milkweed, Asclepias speciosa Torrey (Apocynaceae). We investigated the relative importance of visual and olfactory cues, and the key floral semiochemical(s) mediating attraction of S. myopaeformis to A. speciosa. In field experiments, inflorescences left exposed or enclosed in cheesecloth bags dyed green induced similar visitation rates by moths, indicating that olfactory cues are attractive. Among the >10 floral odourants that elicited responses from moth antennae in coupled gas chromatographic‐electroantennographic detection analyses, phenylacetaldehyde induced the most frequent proboscis extension reflexes of male and female moths. Among eight floral odourants that were field‐tested singly, phenylacetaldehyde attracted 35 times more male and female moths than any other candidate semiochemical. Attractiveness of phenylacetaldehyde could not be enhanced by admixture with other floral odourants at the ratios or concentrations tested indicating that it alone may mediate attraction of S. myopaeformis to the inflorescences of A. speciosa. The potential use of phenylacetaldehyde as bait to monitor or mass‐trap populations of male and female S. myopaeformis should be investigated. 相似文献

Using PD325901 as a starting point for identifying novel allosteric MEK inhibitors with high cell potency and long-lasting target inhibition in vivo, truncation of its hydroxamic ester headgroup was combined with incorporation of alkyl and aryl ethers at the neighboring ring position. Whereas alkoxy side chains did not yield sufficient levels of cell potency, specifically substituted aryloxy groups allowed for high enzymatic and cellular potencies. Sulfamide 28 was identified as a highly potent MEK inhibitor with nanomolar cell potency against B-RAF (V600E) as well as Ras-mutated cell lines, high metabolic stability and resulting long half-lives. It was efficacious against B-RAF as well as K-Ras driven xenograft models and showed—despite being orally bioavailable and not a P-glycoprotein substrate—much lower brain/plasma exposure ratios than PD325901. 相似文献

Single-channel current–voltage (IV) curves of human large-conductance, voltage- and Ca2+-activated K+ (BK) channels are quite linear in 150 mM KCl. In the presence of Ca2+ and/or Mg2+, they show a negative slope conductance at high positive potentials. This is generally explained by a Ca2+/Mg2+ block as by Geng et al. (2013. J. Gen. Physiol. http://dx.doi.org/10.1085/jgp.201210955) in this issue. Here, we basically support this finding but add a refinement: the analysis of the open-channel noise by means of β distributions reveals what would be found if measurements were done with an amplifier of sufficient temporal resolution (10 MHz), namely that the block by 2.5 mM Ca2+ and 2.5 mM Mg2+ per se would only cause a saturating curve up to +160 mV. Further bending down requires the involvement of a second process related to flickering in the microsecond range. This flickering is hardly affected by the presence or absence of Ca2+/Mg2+. In contrast to the experiments reported here, previous experiments in BK channels (Schroeder and Hansen. 2007. J. Gen. Physiol. http://dx.doi.org/10.1085/jgp.200709802) showed saturating IV curves already in the absence of Ca2+/Mg2+. The reason for this discrepancy could not be identified so far. Aims
Along a gradient of diminishing heavy metal (HM) concentrations formed by local inclusions of uranium mine soils into non-contaminated cropland, duplicate 1-m2 plots of 3 winter wheat cvs. (Akteur E, Brilliant A, and Bussard E) were established at 3 positions within a winter rye (cv. Visello) culture. It was the goal to determine permissible soil HM concentrations tolerated by cereal cvs. with variable excluder properties, and regulatory mechanisms which optimize the concentrations of essential minerals and radionuclide analogues in viable seeds from geologically related soils with diverging HM content.Methods
Total metal concentrations / nitrogen species in soils, shoots, and mature grains were determined by ICP-MS / spectrophotometry, and Kjeldahl analyses.Results
No non-permissible concentrations in grains of the 4 cereal cvs. were caused by elevated but aged total soil resources (mg kg-1 DW) in As (156); Cu (283); Mn (2,130); Pb (150); and in Zn (3,005) in the case of Bussard although CdCuZn elicited phytotoxicity symptoms. Uranium (41) contaminated grains of Akteur and Brilliant but not of Bussard and Visello due to their excluder properties. The concentration in Cd (41) had to be reduced to 20/2 mg kg-1 for the production by excluder cvs. of fodder/food grains. Cultivars excluding both HM and radionuclide analogues such as BaCsSr synchronously were not identified. Whereas plant tissue concentrations in the metalloprotein-associated elements CdCoCuMnNiZn rise and fall generally with Norg, grains of the wheat cvs. differed too little in Norg to designate variations in their metal acquisition rates solely as protein-regulated. Wheat grains confined nevertheless the concentrations in Cu to 11–14 mg kg-1 although the respective soil concentrations varied by factor 19. Grain deposition in CaFeMn(Zn) and in nuclides followed the same rules.Conclusions
It is hypothesized that cereals down-/up-regulate grain:soil transfer rates from soils with excessive/deficient trace metal resources to equip viable seeds with an optimum but not maximum in essential minerals.
